Independent Control of Oxygen Concentration for Cell Culture in an Add-on Platform for Multi-well Plates

Oxygen is a key modulator of cellular pathways, but current techniques for probing this important variable lack precise control and efficiency. A microfabricated polydimethylsiloxane (PDMS) insert for multiwell plates has been developed to provide spatial and temporal control over the gas concentration in each well independent of the global incubator condition. Human dermal fibroblast adult cells were used as a cellular model and demonstrated that standard cell lines can be cultured and their behavior modified through oxygen delivery with our platform. The platform can be used to achieve high throughput and novel experimental possibilities in a simple to use format.
